Two structural features account for the prolonged half-life of semaglutide. A modified amino acid at position 8 resists cleavage by dipeptidyl peptidase-4, the enzyme that rapidly degrades native GLP-1. A fatty diacid side chain binds serum albumin, which limits renal clearance and protects the peptide from enzymatic breakdown. These modifications yield a plasma half-life of approximately one week in humans, allowing once-weekly administration. The relationship between plasma concentration and clinical effect varies between individuals, and sources of that variability are still being characterized.
Semaglutide is a synthetic peptide analog of glucagon-like peptide-1 (GLP-1), a hormone released from intestinal L-cells after food intake. The compound belongs to the incretin mimetic class and acts at GLP-1 receptors distributed across pancreatic, gastrointestinal, cardiovascular, and central nervous system tissues. Compared with native GLP-1, the molecule carries structural changes that extend its activity from minutes to roughly one week. It is studied for glycemic control in type 2 diabetes and for weight management, and its effects on cardiovascular and other outcomes remain active research areas.
Receptor binding triggers G protein signaling that raises intracellular cyclic AMP in pancreatic beta cells. Insulin release follows in a glucose-dependent manner, so secretion increases when blood glucose is elevated and diminishes when it is not. The same signaling suppresses glucagon release from alpha cells and slows gastric emptying, which blunts the post-meal glucose rise. In the brain, receptor activation in regions such as the arcuate nucleus is associated with reduced appetite and lower energy intake. How much each of these effects contributes to overall weight change is not fully settled.

Stability studies focus on deamidation of asparagine and glutamine residues, oxidation of methionine, and aggregation into higher-order species. The fatty acid side chain adds susceptibility to oxidative change and can promote self-association at high concentration. Lyophilised material is comparatively robust when kept cold and dry, while aqueous solutions require refrigeration and protection from light. Forced degradation experiments under heat, acid, base, and peroxide conditions establish the specificity of each analytical method. Which degradation route dominates under real storage conditions depends on the formulation and stays formulation-specific.
Handling guidance for research quantities calls for single-use aliquots, an inert atmosphere where practical, and avoidance of repeated freeze-thaw cycles that accelerate aggregation. Certificates of analysis typically report purity by peak area, water content, counter-ion identity, and residual solvent levels. In the scientific literature the compound is usually described by its full amino acid sequence, its registry number, or its structural class rather than by any proprietary label. Reporting standards vary between journals, and reviewers increasingly request raw chromatograms alongside tabulated purity figures. Whether current purity thresholds are adequate for every experimental context is debated.
Reversed-phase high-performance liquid chromatography with ultraviolet detection is the dominant approach for peptide purity assessment, usually paired with mass spectrometry to confirm molecular mass and sequence. Peptide mapping by enzymatic digestion and tandem mass spectrometry locates modifications such as deamidation and oxidation. Quantitation in plasma matrices can be performed by LC-MS/MS after solid-phase extraction. Method validation follows general guidance on accuracy, precision, linearity, and limits of detection. Comparability of results between laboratories, when no shared reference standard is available, remains an open question.
Lyophilised material appears as a white to off-white cake or powder that is hygroscopic, and containers are usually equilibrated to room temperature before opening to limit condensation. Dissolution is performed in water, phosphate-buffered saline, or a mildly alkaline buffer, since solubility rises above neutral pH. Gentle inversion or low-speed mixing is preferred, because vigorous vortexing can promote surface denaturation and aggregation. Complete dissolution may require several minutes, and brief sonication is sometimes applied. Passing the solution through a 0.22 micrometre membrane removes particulates but does not by itself sterilise the liquid.
Storage at minus 20 degrees Celsius or lower in a desiccated container preserves the peptide for extended periods, while working solutions are commonly held at two to eight degrees Celsius for short intervals. Light exposure and repeated freeze-thaw cycles accelerate degradation, so dividing material into single-use aliquots is generally recommended. Adsorption to glass and plastic surfaces can lower the measured concentration of dilute solutions, particularly below one milligram per millilitre. The degradation routes most often reported for GLP-1 analogues are deamidation, methionine oxidation, and backbone hydrolysis. Relative rates under specific conditions are frequently described only for individual formulations.
Semaglutide is a synthetic peptide analog of human glucagon-like peptide-1, developed by Novo Nordisk and first approved in 2017 for type 2 diabetes. It belongs to the incretin mimetic class, a group of agents that reproduce the glucose-dependent actions of endogenous GLP-1. The molecule was engineered to resist degradation by dipeptidyl peptidase-4 and to bind serum albumin, extending its half-life from minutes to roughly one week. Approval for chronic weight management followed in 2021, based on large cardiovascular and obesity outcome trials.
GLP-1 receptors are expressed on pancreatic beta cells, in the gut, and in several brain regions. Receptor activation raises cyclic AMP, enhances glucose-dependent insulin secretion, and suppresses glucagon release when blood glucose is high. Effects on gastric emptying and on hypothalamic appetite circuits reduce energy intake. Because insulin release remains glucose-dependent, the risk of hypoglycemia is low when the drug is used alone. The precise contribution of each pathway to body weight change in humans remains an area of active investigation.
Clinical studies of semaglutide generally measure glycated hemoglobin, fasting plasma glucose, body weight, and composite cardiovascular endpoints. The SUSTAIN program enrolled adults with type 2 diabetes, while the STEP program focused on obesity without diabetes. Administration follows a stepwise escalation schedule designed to limit gastrointestinal effects during the first weeks. Reported outcomes include mean percentage weight change, the proportion of participants reaching defined weight-loss thresholds, and rates of nausea, vomiting, and diarrhea. Long-term data on durability after treatment stops are still limited and remain a topic of ongoing research.
Receptor activation raises intracellular cyclic AMP through Gs coupling, which promotes glucose-dependent insulin release and suppresses glucagon secretion when blood glucose is elevated. Effects outside the pancreas include slower gastric emptying and altered appetite signalling in the hypothalamus and hindbrain. The relative contribution of each tissue to overall metabolic outcomes remains an area of active investigation. Central mechanisms in particular are inferred mainly from animal models and indirect human measures rather than direct observation.
Serum protein binding dominates the pharmacokinetic profile. The attached chain associates strongly with albumin, shielding the peptide from enzymatic attack and slowing filtration by the kidney. This interaction extends the circulation half-life to roughly one week in humans, which supports weekly administration intervals. An oral version pairs the peptide with an absorption enhancer that transiently alters gastric epithelium, permitting limited uptake; bioavailability by that route is substantially lower than by injection.

==== United States ==== In March 2020, United States President Donald Trump announced that remdesivir was available for "compassionate use" for people with COVID‑19; FDA Commissioner Stephen Hahn confirmed the statement at the same press conference. It was later revealed that Gilead had been providing remdesivir in response to compassionate use requests since 25 January. On 23 March 2020, Gilead suspended access for compassionate use (excepting cases of critically ill children and pregnant women), due to short supply, citing the need to continue to provide it for testing in clinical trials. In May 2020, the US Food and Drug Administration granted Gilead emergency use authorization (EUA) for remdesivir to be distributed and used by licensed healthcare providers to treat adults and children hospitalized with severe COVID‑19. Severe COVID‑19 is defined as patients with an oxygen saturation (SpO2) <= 94% on room air or requiring supplemental oxygen or requiring mechanical ventilation or requiring extracorporeal membrane oxygenation (ECMO), a heart–lung bypass machine. Distribution of remdesivir under the EUA was controlled by the US government for use consistent with the terms and conditions of the EUA. Gilead supplied remdesivir to authorized distributors, or directly to a US government agency, who distributed it to hospitals and other healthcare facilities as directed by the US government, in collaboration with state and local government authorities, as needed.
For most of the history of wine, winemakers did not know the mechanism that somehow converted sugary grape juice into alcoholic wine. They could observe the fermentation process which was often described as "boiling", "seething" or the wine being "troubled" due to release of carbon dioxide that gave the wine a frothy, bubbling appearance. This history is preserved in the etymology of the word "yeast" itself which essentially means "to boil". In the mid-19th century, the French scientist Louis Pasteur was tasked by the French government to study what made some wines spoil. His work, which would later lead to Pasteur being considered one of the "Fathers of Microbiology", would uncover the connection between microscopic yeast cells and the process of the fermentation. It was Pasteur who discovered that yeast converted sugars in the must into alcohol and carbon dioxide, though the exact mechanisms of how the yeast would accomplish this task was not discovered till the 20th century with the Embden–Meyerhof–Parnas pathway. The yeast species commonly known as Saccharomyces cerevisiae was first identified in late 19th century enology text as Saccharomyces ellipsoideus due to the elliptical (as opposed to circular) shape of the cells. Throughout the 20th century, more than 700 different strains of Saccharomyces cerevisiae were identified. The differences between the vast majority of these strains are mostly minor, though individual winemakers will develop a preference for particular strains when making certain wines or working with particular grape varieties.
=== Ketoacidosis === Ketoacidosis is a pathological state of uncontrolled production of ketones that results in a metabolic acidosis, with serum ketone levels typically in excess of 3 mM. Ketoacidosis is most commonly caused by a deficiency of insulin in type 1 diabetes or late stage type 2 diabetes but can also be the result of chronic heavy alcohol use, salicylate poisoning, or isopropyl alcohol ingestion. Ketoacidosis causes significant metabolic derangements and is a life-threatening medical emergency. Ketoacidosis is distinct from physiological ketosis as it requires failure of the normal regulation of ketone body production.
== Non-enzymatic processes == Protein backbones are very stable in water at neutral pH and room temperature, although the rate of hydrolysis of different peptide bonds can vary. The half-life of a peptide bond under normal conditions can range from 7 years to 350 years, even higher for peptides protected by modified terminus or within the protein interior. The rate of hydrolysis however can be significantly increased by extremes of pH and heat. Spontaneous cleavage of proteins may also involve catalysis by zinc on serine and threonine. Strong mineral acids can readily hydrolyse the peptide bonds in a protein (acid hydrolysis). The standard way to hydrolyze a protein or peptide into its constituent amino acids for analysis is to heat it to 105 °C for around 24 hours in 6M hydrochloric acid. However, some proteins are resistant to acid hydrolysis. One well-known example is ribonuclease A, which can be purified by treating crude extracts with hot sulfuric acid so that other proteins become degraded while ribonuclease A is left intact. Certain chemicals cause proteolysis only after specific residues, and these can be used to selectively break down a protein into smaller polypeptides for laboratory analysis. For example, cyanogen bromide cleaves the peptide bond after a methionine. Similar methods may be used to specifically cleave tryptophanyl, aspartyl, cysteinyl, and asparaginyl peptide bonds. Acids such as trifluoroacetic acid and formic acid may be used for cleavage. Like other biomolecules, proteins can also be broken down by high heat alone.

20,000, 11 December 2007- On 29 October 2007, the SENSEX crossed the 20,000 mark for the first time with a massive 734.5-point gain, but closed below the 20,000 mark. The SENSEX would close above the 20,000 mark for the first time on 11 December 2007. It took 42 days after reaching the 19,000 milestone to close above 20,000 points for the first time. The journey of the last 10,000 points was covered in just 483 sessions, compared to 7,297 sessions taken to touch the 10,000 mark from its base value of 100 points. In the second half of 2007 alone, the SENSEX reached six 1,000-point milestones.
There are bacteria combining these two ways of nutrition: green sulfur bacteria and purple sulfur bacteria. Also sulfur-oxidizing bacteria can go into symbiosis with larger organisms, enabling the later to use hydrogen sulfide as food to be oxidized. Example: the giant tube worm. There are sulfate-reducing bacteria, that, by contrast, "breathe sulfate" instead of oxygen. They use organic compounds or molecular hydrogen as the energy source. They use sulfur as the electron acceptor, and reduce various oxidized sulfur compounds back into sulfide, often into hydrogen sulfide. They can grow on other partially oxidized sulfur compounds (e.g. thiosulfates, thionates, polysulfides, sulfites). There are studies pointing that many deposits of native sulfur in places that were the bottom of the ancient oceans have biological origin. These studies indicate that this native sulfur have been obtained through biological activity, but what is responsible for that (sulfur-oxidizing bacteria or sulfate-reducing bacteria) is still unknown for sure. Sulfur is absorbed by plants roots from soil as sulfate and transported as a phosphate ester. Sulfate is reduced to sulfide via sulfite before it is incorporated into cysteine and other organosulfur compounds.
The classic model for the enzyme-substrate interaction is the induced fit model. This model proposes that the initial interaction between enzyme and substrate is relatively weak, but that these weak interactions rapidly induce conformational changes in the enzyme that strengthen binding. The advantages of the induced fit mechanism arise due to the stabilizing effect of strong enzyme binding. There are two mechanisms of substrate binding: uniform binding, which has strong substrate binding, and differential binding, which has strong transition state binding. The stabilizing effect of uniform binding increases both substrate and transition state binding affinity, while differential binding increases only transition state binding affinity. Both are used by enzymes and have been evolutionarily chosen to minimize the activation energy of the reaction. Enzymes that are saturated, that is, have a high affinity substrate binding, require differential binding to reduce the energy of activation, whereas small substrate unbound enzymes may use either differential or uniform binding. These effects have led to most proteins using the differential binding mechanism to reduce the energy of activation, so most substrates have high affinity for the enzyme while in the transition state. Differential binding is carried out by the induced fit mechanism – the substrate first binds weakly, then the enzyme changes conformation increasing the affinity to the transition state and stabilizing it, so reducing the activation energy to reach it.

=== Prevention === A migraine management plan often includes lifestyle modifications to cope with migraine triggers and reduce the impact of comorbidities. Recommended lifestyle modifications support a consistent lifestyle, through regular sleep patterns, regular eating, staying hydrated, managing stress, engaging in moderate exercise, and maintaining a healthy body weight. Avoiding dietary triggers and caffeine overuse may also be recommended. Improving sleep patterns may be particularly helpful in reducing migraine frequency for adults with chronic migraines. Behavioral techniques that have been used in the treatment of migraines include Cognitive Behavioral Therapy (CBT), relaxation training, biofeedback, acceptance and commitment therapy (ACT), as well as mindfulness-based therapies. These treatments can reduce migraine frequency both on their own and in combination with other treatment options. For children and adolescents, CBT and biofeedback strategies are effective in decreasing the frequency and intensity of migraines. These techniques often include relaxation methods and promotion of long-term management without medication side effects, which is emphasized for younger individuals. A variety of possible diets have been proposed, including ketogenic diet, Mediterranean diet, DASH diet, and high intakes of fruits, vegetables, legumes, and oil seeds. Evidence supporting interventions such as transcranial magnetic stimulation and transcutaneous supraorbital nerve stimulation remains limited.
===== Pharmacodynamics ===== Cyproterone acetate suppresses libido by directly reducing the level of active androgen, testosterone, in males. The suppression of testosterone level results from its inhibition of the release of luteinizing hormone (LH) from the anterior section of the pituitary gland, interfering with testosterone production from the testes as LH stimulates testosterone production. It also blocks the conversion of testosterone to dihydrotestosterone for action. In addition to this mechanism, it also competes for the androgen receptors against testosterone and dihydrotestosterone, causing interference with the androgen-receptor interaction on the reproductive organ, thereby lowering sexual desire.
== Use and effects == In his book PiHKAL (Phenethylamines I Have Known and Loved), Alexander Shulgin lists TMA-2's dose as 20 to 40 mg orally and its duration as 8 to 12 hours. In earlier publications, it was described that threshold effects occur at a dose of 10 mg orally, an effective dose is 16 to 20 mg orally, its onset of psychoactive effects is after 1 hour, and a plateau of effects occurs from 3 to 6 hours following administration. The drug is much more potent than its positional isomer 3,4,5-trimethoxyamphetamine (3,4,5-TMA, TMA, or TMA-1), which is said to be active at doses of 100 to 250 mg orally and to have a duration of 6 to 8 hours. However, DOM (2,5-dimethoxy-4-methylamphetamine), the analogue of TMA-2 in which its 4-methoxy group has been replaced with a more lipophilic 4-methyl group, is about 10 times more potent than TMA-2. TMA-2 has been said to have a sharp dose–response curve, with several additional toxic symptoms occurring at doses of 25 to 30 mg. The effects of TMA-2 have been reported to include color and contrast enhancement, closed-eye imagery like kaleidoscopic images, visuals such as visual distortion and movement, auditory enhancement, increased salience of objects in one's environment, cosmic thinking, time dilation, and music and erotic enhancement.

Native GLP-1 is degraded within minutes by dipeptidyl peptidase-4 and cleared quickly. Semaglutide carries a position 8 substitution that blocks that cleavage and a fatty diacid chain that binds albumin. Together these changes extend its circulating half-life to about one week.
